The difference in dissolved oxygen (DO) between the initial measurement and the fifth day measurement represents the biochemical oxygen demand(BOD).

I used to get an empirical equation between COD and BOD for certain wastewater. This equation differ from waste to another.

Bernice, you should get a copy of Standard Methods for the Examination of Water and Wastewater, published by APHA, AWWA, and WEP organisationsA. They have the methodology for measuring BOD documented very well. As Avinish stated above, the difference in the zero and 5 day reading is the BOD5, but you may need to dilute the water, or add a seed water, or do some other process to get a reading that is accurate.
