Although freshly prepared substrate may enhance you obtaining good result of activity but as stated earlier by other colleagues, you can give it a trial by running with both freshly prepared substrate and prepared and stored before use .Then, compare the results.

To refresh the substrate must be

washing two or three times with wash solutions depend on type subtrate. After that you can do purification correctly.

Azocasein solution as a substrate is stable for 3 months if kept at 4 cent.previously i did work with and that was what i found.
