I am currently working on Vibrio parahaemolyticus from seafood. For the isolation of it I am using both TCBS (Thiosulfate citrate bile salts sucrose agar) and Chromagar Vibrio. I have noticed that quite a lot of typical colonies on TCBS that are also positive on Chromagar Vibrio exhibit a quite hard colony, making lift of the colony for subculturing quite difficult and often unsuccessful. This colony morphology has not been observed in other media (Chromagar Vibrio, T1N1) but solely on TCBS. Since I do not have much experience on Vibrio, is this common or there is something going wrong with my TCBS plates?
