I'm getting some tissues from a collaborator who already fixed them (mouse lymph nodes) in formaldehyde and then transferred to 70% ethanol for storage. Normally I would take fixed tissues into 30% sucrose before embedding in OCT for cryosectioning. Does anyone know if the 70% ethanol step will disrupt this? The colleague normally takes them into paraffin, but I prefer doing frozen sections. We want to do IHC on the sections.

I think that is a bit tricky. I would imagine that the tissues would be a bit dehyrated in ethanol and if you cryocut them as they are they are likely to crack. I would try to rehyrdrate first in PBS for a few hours to wash ethanol off, and then do your usual overnight 30% sucrose before embedding in OCT.
