I'm getting some tissues from a collaborator who already fixed them (mouse lymph nodes) in formaldehyde and then transferred to 70% ethanol for storage. Normally I would take fixed tissues into 30% sucrose before embedding in OCT for cryosectioning. Does anyone know if the 70% ethanol step will disrupt this? The colleague normally takes them into paraffin, but I prefer doing frozen sections. We want to do IHC on the sections.
