Could someone inform me if my thinking is correct about the utilization of the BAM Most Probable Number (MPN) method for quantification? I am quantifying Salmonella in some GI content using the BAM MPN Excel file from the FDA.
I have an original sample of 0.20 grams which I have diluted in 2 mL of PBS (0.1g/mL concentration now). I am performing 8, 10-fold dilutions of this sample (100 uL sample into a now total 1 mL broth). Since this method quantifies based on MPN/g, do I start by saying 0.2 grams, 0.02, 0.002...etc or use the 0.1g/mL?
I think perhaps I did not need to perform the initial sample dilution of 1/10.
