Thermo recommends no DTT in cobalt chromatography (Hispure, # 89964) buffers. I wonder if 5mM DTT going to ruin my cobalt chromatography purification?Can anybody help me with this?
I believe you are doing the classical His tag purification. Such His tag purification need to avoid DTT. However, new column such as iMac from GE Healthcare could resist to DTT stripping more (see GE high bounds columns or IMAC colomn from Pierce). The high quality standard now for His tag purification is to use low reduction buffer such as TCEP.
