Hello All,

I recently retrieved some total RNA-seq data from mouse and I'm having trouble interpreting some of the QC figures. Most samples are as expected, however one of the samples (sample 5 in the attached image), appeared to have skewed gene body coverage.

Samples were prepared using Illumina total RNA kit, and run on the NextSeq, paired-end. The sample did show poor rRNA depletion (I used an rRNA depletion kit on all samples), but others showed this too and did not show quite the dramatic skewing that is seen here. The gene body coverage heatmap (part of which is attached) was generated using http://rseqc.sourceforge.net/  geneBody_coverage.py

If anybody has any idea as to what this might be showing, I'd be really grateful for any input.
