specific primers were used and found another technique by serologic method was performed using different antisera and this is simple and the result not specific but very cheap, in another hand, I suggest PCR for the confirmative results
Thanks for your answer. Please refer to my profile and you can see that I used SBT method in most of my projects and cytotoxicity test in another project. The aim of my question was to compare between the methods used in HLA typing and look for the most accurate method.
Direct sequencing using any of the available sequencing platforms such as NGS would offer the best means of determining the HLA type, in my opinion. The advantages of this method include being accurate, sensitive, high throughput, and ability to discover novel HLA types. Also, unlike the PCR-based methods, it does not require prior knowledge of the gene sequence to determine. Disadvantages are the cost, availability of the technology, know-how, and may be tedious and require rigorous bioinformatics input to accurately determine the HLA type.
