in the biosynthesis of silver nanoparticles from plant by using 1mM AgNO3 two peaks at 668 and 330 nm had observed. at 0ml of plant extract 668 nm had be observed . i m confused about which peak should be considered for plant synthesized silver nanoparticle . in this assay the reference should be 1mM AgNO3 solution or distilled water ?
Silver nanoparticles appear yellowish brown color in aqueous medium. As the different leaf extracts were added to aqueous silver nitrate solution, the color of the aqueous solution changed from faint light to finally brown indicating AgNP formation. Absorption spectra of Silver nanoparticles formed in media has absorption peak in the range of 425 to 475 nm.

The absorption sectra of the samples were taken 340 – 540 nm using UV - spectrophotometer
Dear Rabia in your case both the peaks does not indicate the AgNPs. May be due to high amount of plant extract suppressed the peak of the AgNps if its there. Secondly Your peak 330 nm indicates AgNps oxidized and no more in media. Moreover I suggest you to prior using plant extract try to add more filtration steps.
And it is fine that you synthesis with green approach and made it non toxic to the environments. Best of luck
Dear Rabia none of the peaks indicate the formation of AgNPs. Depending upon the size of your nanoparticles the SPR peak ranges between 420 - 450 nm. Conduct experiments by varying the dose of the extract. Formation of AgNP is indicated by a characteristic brown colour colloidal mixture.
i have taken the extract 1 to 10 ml and adding 1 ml of AgNO3 in each after 24, 48, 72 hours directly spectrophotometric readings had be taken ,,,,,,but the results are same, is there any method for the purification of AgNps for the spectrophotometric study
Use dilute concentration of extract. You will get peak at desired position 420 to 450nm
i myself syntheiszed silver nanopartilces by plant extracts and range was coming between 420-430 nm.

We got at 448 nm, confirmed by characterization studies like dynamic light scattering
