It is better to first extract the plant sample with an extractant (e.g., buffer, organic solvent, detergent, etc.) and apply the extract to the column. If you apply the dry material to the column, during the elution, the substance(s) you are trying to isolate will continually leach or "bleed" from the sample into the column eluent, so little to no separation of the components will occur.
You may have to try several different extractants to find the one that works the best. If you use an organic solvent as an extractant, be sure that it is compatible with the resin used in the column.
