Hi, everybody!
shRNA can use to knock down the protein expression. I want to knock down two proteins‘s expression together in vivo. Is it possible if I design a virus like AAV-GFP-shRNA1-shRNA2? Can it work? Or it is better to inject two viruses to knock down them,but in this case I am thinking there might be some cell which only got one virus expressed.

You need to construct a plasmid which has either both shRNAs and each shRNA has its own promoter separately or both shRNAs, separated by a linker, regulated by one promoter. I have done similar thing "silenced 2 genes at the same time", but used CRISPRi-Cas9 system and worked. Hope, it is going to work for you .

just clone one shRNA under human U6 promoter and another under mouse U6 promoter in the same vector.
Or you can use H1 promoters. This will work 100%
