If I am performing an experiment in which plants are exposed to salt stress (eg. 0 and 100 mM) at both in vitro and ex Vitro conditions, do I need to perform one -way ANOVA or two-way ANOVA to get the difference between the traits (eg. plant height, FW, TWC), treatment and method?.
Are the same plants exposed in invotro and ex vitro conditions or is there a group of plants for each condition? Are the same plant fist exposed to no salt and then salt stress?
Neverthelesse, you have to factors, stress (no salt vs salt) and condition (in vitro vs ex vitro), so a two way ANOVA sounds fine. But you have to consider if each factor is a repeated measures independent factor, to choose the correct model. Additionally, you should always check the assumptions for each model!
The Two-way ANOVA for ex-vitro and in-vitro condition analysis is appropriate, and I encourage you to perform a post-test such as Tukey or Bonferroni (if your are using several salt concentrations, exposition times, plant species or varieties).
