look up the properties of the compound see what solvents they're soluble in, it's possible they may be difficult to analysis by lc ms ms, gc ms ms may be your only choice.
Your compound sounds very lipophilic. Are you using a C18 or C8 HPLC column or are you direct injecting the material without a column? If the material is very lipophilic, it may not elute from C18 or C8 without tetrahydrofuan (THF) in your organic mobile phase (for lipids 20-40% THF in ACN is used for lipid analyses).
If your compound is that non-polar, you may be able to dissolve in THF and dilute slightly with water (50% max) before you analysis. If your compound elutes early on these columns, then I would try isopropanol instead of THF.
