I use FDA method for that analysis. I have standard curve, soil moisture content (%) and reading (OD 490) and I use 2 g of wet soil and 2 replicates for each sample.Thanks.
Calculate the fluorescein concentration from the calibration curve. Substrate the control reading (hydrolysis in absence of soil) from the sample reading (hydrolysis in presence of soil) and express activity as " micro-gram of released fluorescein per gram soil dry mass over hour" using the following formula:
Micro-gram Fluorescein/ (g dry soil x h) = Sample - Control / Ws x Ds
Sample = pNP concentration of the soil sample (micro-gram)
Control = pNP concentration of the Control ( micro-gram)
I have a treatment contain enriched compost with microbes. I need to evaluate the microbes but don't want to proceed a CFU. Can I use the t60 uv vis spectrophotometer ?. And if yes what procedure to follow
