For surface roughness analysis of PDMS surface, you can consider by digital microscopy or AFM. Mostly AFm is preferable for Ra, Rz and Rq etc roughness parameter evaluations. if the PDMS is soft surface then you might need to consider for some sample preparation beforehand for AFM for competitive results. Hopefully you can get your results. if anything else, let us discuss.

3D surface profiler can also give you information about surface roughness if you don't have AFM technique.

AFM. To induce a specific roughness try using a substrate in the same range.
