I have used 30nM and 100nM to stimulate PMNs and it worked quite well esp. for 100nM. But I personally focused on measuring mRNA and protein levels of pro-inflammatory cytokines, so I used longer time points that was 6 and 12 hours respectively. Off course I dealed with quite high variability between the donors.
Additionally, I have checked activation of these cells by measuring Ca flux and even 10nM of fMLP was sufficient to induce Ca flux (that means activating the receptors of fMLP on these cells) but this info might not be so relevant to you.
