I have sulfo tagged an antibody. The incorporation ratio is 10 but the MSD signal is low (in 3 digits). What could be the possible causes?

I have sulfo tagged an antibody two times. The first time my incorporation ratio was 2. The second time we added sodium bicarbonate and the incorporation increased to 10. However when I did my assay on MSD, the max signal was 250 RLUs on the top standard conc of 0.5mg/ml.

What can cause this to happen?
