I need to determine the quantity of Trichoderma conidia powder to be applied to a set quantity of vermicompost. what method can i use to determine this. how can i prove that the quantity of powder is the best for application?
Hi Dr. Matny, so i did a serial dilution with one gram of the powder and used the hemocytometer to count the spores of the 10^-3 dilution. I got 1.0 x 10^6.21 spores. How do i proceed from here?
with hemocytometer method you are counting the active and non active spores.
A total viable spore count method is counting a life spores, take1g of the Trichoderma powder in 9ml of sterilize water, mix it well this is well be 10 ^ -1and make dilution 10 ^ 2 , 10 ^ -3, 10 ^ -4.......10 ^ -7 , and you take 1ml of the final dilution and put it on 9cm dim petridish and pure PDA medium on it with mixing. Put it in 25C for 2 day and count the colony that you get and multiply it with the upset of the dilution you will get the spore concentration of life active spores.
You can have different conc. of conidia per Gram vermicompost and observe the effect on different growth parameters. It will help you in finding out effective dose.
