I have ZnO and MgO nanoparticles? I want to know that can I measure their concentration using ImageJ? I have their confocal images. I have seen papers on cell counting and DNA concentration measurements using ImageJ but not with nanoparticles. What if I have mixed nanoparticles samples then how can I measure their concentration using ImageJ? or if ImageJ is not appropriate what method should I use to estimate concentration?
This depends on your images, but sounds possible. If you want overall concentration, you will need to threshold the images first (I think it’s in Image > Adjust > Threshold), then measure the area of the thresholded mask compared to the area of the entire image. No. of cells can be counted and even amount of DNA also can be determined. But how will you measure the concentration of NP. Its not possible Dr. The programme will provide you the intensity of images in different modes. Even if you denote Y axis with intensity, what will be there in the X?
In principle if you want to calculate the concentration of nanoparticles using an image you would need to do using a calculation like the one used to count cell with an hemocytometer (https://www.nexcelom.com/how-to-count-cells-using-a-hemacytometer/). The problem is that you need to know the volume used since that could increase your concentration, since that volume would give you an idea of your concentration after counting your nanoparticles. Maybe you can adapt from the hemacytometer model to something that fits your needs.
