When you plan to use glass bottom dish or coverslips, you also need to pay especial attention to the glass quality. You need to pre-treat the glass with acid overnight before coating. The acid will help to make the glass surface a little rough which may facilitate adherence much more strongly. The acid will also remove any trace amounts of impurities that may interfere with adherence.
Acid washing of coverslips is recommended particularly if you are planning to grow adherent cells on the surface and it helps polypeptides to adhere to the glass. Since it is a long protocol, you need to plan in such a way that you make a batch of acid-washed coverslips and store them in a clean container to be used later for your experiments.
The protocol will include the following steps:
1. Cleaning slides and coverslips
Washing with PBS/water
Acid Washing
2. Sterilizing coverslips
Using Ethanol, UV Light or Autoclaving
3. Coating slides and coverslips
Poly-Lysine, Collagen, Fibronectin, Laminin
