I am formulated herbal ointment, and screening o0f wound healing activity, in literature most of them used povidone iodine as a standard. But I am studying herbal formulation, which is the suitable standard herbal ointment
Yes there are many herbal formulation is present for making the standard. You can take the help of good Journals for getting Clear idea about your queries.
I am afraid you can not take any herbal formulation as a standard for your aim. They contains different compounds with different chemical nature. Because almost all researches like yours for creating herbal ointment for wound healing compared their results with standard substances (i.e. povidone iodine, Nitrofurazone etc.). I hope it is enough to compare your results with their activities.
Generally, known single effective component is used as a control at a fixed concentration.
As phytochemical concentration of plant parts may vary with soil of cultivation, maturity level of plant, climate etc., it is very difficult to fulfil such criteria by any multi- component herbal ointment.
I agree with already existing comments. Get a known (even if synthetic) chemical compound with wound healing activities, prepare different concentrations and compare the activity of your formulation.
Many ointments preparations for wound healing are available, especially with calendula, marshmallow, or Echinacea herbs as active ingredients, you may try them caparatively.
The main problem of natural plant origin ointments is what single compound is responsible for the shown biological activity (wound healling) here. How do you ensure your ointment activity comparing with another ointment's effect (as you take as a standard) if you do not know what compounds are there and their conc. in it?
Further to the answer given by Nanloh Samuel Jimam, comfrey and the plantago species. I've made great ointments using dried comfrey, olive oil and a small amount of beeswax.
In my experience, herbs never have just one single compound responsible for their action. Sure you might find one compound that appears to do the main work but then there are others to also assist and still more that prevent possible side effects.
Just look at turmeric. The suggestion that curcumin is the only active compound has now been overturned as more and more compounds are found to have beneficial effects.
There are many research articles published in standard ayurveda journal with proven significant effect of wound healing property of herbal drugs, you can refer those as standard...

The main aim is to compare the pharmacological activity of test drug with povidone iodine as standard then you may proceed with povidone iodine. However many herbal ointments are available in the market and we can use those as standard.
