hello, i got a problem in the adipogenesis.
The human MSCs were differentiated into adipocte. 10days later, the mutant MSCs and the WT MSCs were red oil stained and QPCR test. But there's a probelm, the mutant MSCs' gene marker of adipogenesis were higher expressed than the WT MSCs', while the red oil staining showed that the phenotype of mutant MSCs didn't show the strong phenotype.So, why did this happen?
Which adipogenic markers did you analyze? We usually detect adiponectin and PPARgamma2 upregulation by qPCR
If I understand correctly, the problem is with the Oil Red staining, right? Which staining protocol do you use?
Also, the working Oil Red solution is only stable for about 1 to 2 hours. Are you preparing a new working solution each time? You can prepare a stock solution for storage and make a working solution from the stock solution each time you need it.
You can also try other stains specific for lipids, like the black Sudan staining.

Thanks for your advice. i usually use the PPAR-gamma, FABP4 and CEBP-alpha as the adipogenic markers. As you said, maybe i can have a try to change the gene markers and test again.
