Hi, can anyone assist me with the acceptable range for peak resolution. Literature states the Rs value should ideally be above 1.5.
From my DOE results I am obtained Rs values in the range 15 - 36 and that is very high, I have done manual calculations and I am still getting very high values >15.
The instrument I am using is Thermoscientific Ultimate 3000 HPLC and the software is Chromeleon 7.1.
Unami Sibanda: HPLC peak Resolution must be calculated from an "INITIAL Peak" (P1) as it is a comparative value to a second peak (P2). If you were to use the "wrong" peak, then you could easily show results with extremely large R values. "Resolution" calculations require that the data used is taken from two different peaks using a properly developed method, showing clean baselines, peaks with proper Gaussian shape, Peak Width measured at half height (50%) for ASTM or USP calculations and proper integration settings. Errors or issues with any of these may result in calculation errors.
