Typically, you strip it with 1M NaCl until the baseline levels off. Then you wash with dH2O until the baseline levels off. Then you equilibrate with your loading buffer until the baseline levels off.

Certain circumstances require more stringent column stripping. If you observe significant reduction in loading capacity, or increased non-specific elution, you may need a separate wash with 1N NaCl to remove sticky stuff.

Before sample application, column washed with resin in 20% ethanol.

After completion of purification , column cleaned with

1. 2M Nacl (High Salt Concentration)- remove ionically bound protein

2. 1M NaOH - remove hydrophobic protein and lipoprotein

3. 70% ethanol - Hydrophobic bound moleculers and lipids

4.Distilled water- Final cleanup to store
