i am working in a project where we want to separate the magneto-tactic bacteria from the Egyptian aquatic environment and use the magnetosomes of these bacteria after that
what is the technique the isolation of these types of bacteria and without using a differentiation media
if there is good and not expensive protocol to separate it
and in which aquatic environment we can find this types of bacteria

Take a Pasteur pipette, fill it with water and put some glass wool between thick and thin parts of pipette. Inoculate thick part of pipette with your sample and place a magnet in front of the closed end of the thin part. MTB should mirgate through the glass wool and concentrate in the thin part.

but i need to know more about the cultivation protocols? and how to increase there number? and the media types to cultivate them ?
and to know more about the oxygen sulfur gradient media which have the optimum conditions for cultivating these bacteria? and to know how to make it ?
I suppose you are not yet specific in your mind what and how you want to do, what you are doing. Thus, I recommend you to discuss with your supervisor, colleagues and read research papers related to your topic. Why not read through the whole original paper mentioned in previous links.

Yet actually I am working on a project about isolating the MTB and extract the magneto-some from it, and use it after that
and i have collected different protocols for the isolation process.
and i am seeking your help to find more information about the Precautions and considerations that I have to take before beginning in the isolation steps of the bacteria.
