I am currently performing enzymatic hydrolysis (ie xylanase, cellulase) on lignocellulosic substrates. Do avoid the reconsumption of sugars by endogenous bacteria naturally present on the LC, a sterilization step is generally applied. Nonetheless such treatments is expensive and modify the structure of the LC.
Another possibility is the addiction of antibiotics but such option is not recommended in the case of further biological processes like anaerobic digestion or dark fermentation using mixed cultures.
Thus, I would like to know if other research group have developped alternative methods to the sterilization step.

I would recommend the exploration of sterilisation by irradiation, possibly UV.

Dr. Nisola we already run a sample without sterilization but the reconsumption is significant.
Dr. Asamoah, good idea the use of irradiation. But i think that the UV irradiation will lead to some structural or chemical modifications ? Nonetheless, it could be interesting to compare this physicochemical changes with classical sterilization.

Florian treatments such as irradiation, heat or chemical would certainly affect chemical structure and/or integrity/nature of lignocellulose. However, minimal amounts which may go as far as influencing conformational changes may be accommodatable since conformational changes in several instances is completely reversible. Only which conformational type in which experiment ought to be accurately known, so as to afford interesting conformational information in relation to hydrolysis.
Do you try hot CO2 for 20 minutes / high pressure hot water pass through the your materials but i don't known your process if possible sent entire process step wise i may try solve your problem
What is the economic objective of the question.
If the objective is to deal with a lignocellulosic biomass waste, then the economics of sterilization will become significant. We have developed a process that converts this biomass into multiple high value products that accept a non-homogenous, non-sterile feedstock and processes it under non-sterile conditions. It works at ambient pressure at about 40 C. Attached is a paper describing the process and the results that have been confirmed by independent laboratories. The paper is scheduled for publication in June in the Journal of Sustainable Development.
