I have to analyze 4 different substrates for larvae rearing, and I made 3 replications (each one a sample of 30 larvae).

I am starting with ANOVA and then post hoc test, like Tukey.

It is unclear for me how I should use the replications :

I can compare the means of the 3 replications (hence a table with 4*3 cells), but then I cannot compare the variance between the 30 larvae of each sample.

Otherwise I added up all replications to form 4 groups (4 treatments) of 90 larvae, but then it's not longer replications, it is just one big group...

Otherwise I could just analyse the variance (ANOVA and TUKEY) for all 3 replications, and see if the results are coherent, but I did not come accross that kind of tables in literature...

Another question (sorry I am really starting with this) : how do I categorize the results with significant letters, like in scientific articles (Like A, B , C) ? Is it juste something that I write myself, if p-value is below my treshold ?

Like, how do I do if let's say A is not significantly higher than B, and B is not significantly higher than C, but A is significantly higher than C ?

I cannot find a clear methodology for that...
