Thanks for sharing your interesting question with the RG community. HClO4 in glacial acetic acid is a very strong acid that can be used to titrate weak bases such as amines.

When titrating Tris (= tris(hydroxymethyl)aminomethane) with HClO4, the amino group is protonated, so the stoichiometry of the reaction should be 1:1. 1ml of a 0.1N HClO4 solution then corresponds to 12.114 mg Tris.

In your case (Tris-HCl) the amino group is already protonated, so I would intuitively assume that the molecule cannot be titrated further in glacial acetic acid. Some additional information or literature on the method you are using would be helpful to give you an answer to your question.

Add Mercuric acetate for intererence of HCl before titration in Tris HCl solution then titrate with perchloric acid 0.1 M/N.

if possible maybe you can try with potentiometer. If tris HCl not completely dissolve, you can stir the sample automatically. Hope it helps
