I am trying to find out how to store sclerotia from Sclerotinia sclerotiorum. I have three isolates from cucumber, sunflower and squash, can anyone advice on how to store this properly if I want to culture it in the future?
Hi Pfano : The best way to preserve the fungi is to keep the most structure that can survive to long term, for the Sclerotinia sclerotiorum the best is to preserve the sclerotia, you can preserve it as dry in room temp for more than 1 year , but if you want to preserve for long term dry the sclerotia and keep it in vial at 4C or -80C. Several methods cited in the book of Saharan, G. S. "Sclerotinia Diseases of Crop Plants: Biology, Ecology and Disease Management". Springer, 2008. have recommended to grow S. sclerotiorum on wheat grain before long-term storage at 4oC.
I agree with all the previous respondents. Simply insure that the sclerotia are dry (put in a 40C oven) and then store as cool as possible.. -4, -20 or -80. Sclerotia are amongst the easiest fungal propagules to store for a long period of time. The larger the sclerotia the longer they will survive.
