I've recently started paraffin sectioning of muscle tissue with some success, however in some sections I find tears in the tissue that all run parallel with each other and I believe run perpendicularly to the motion of the blade.
I've tried frequently changing the blade so its not dull, I've watched to keep my blocks cold and still the problem persists.
I've fixed my TA mouse muscle in 10% formalin for 24 and 48 hours so i dont think they have been underfixed and paraffin embedding was done in histology core so I dont believe that is the issue.
Could this be caused by too great of a blade angle? Ive been using between 8-10 degrees
