I am having Alpha tocopheryl acetate but want to calculate only Alpha tocopherol content from it without using any conversion factor. By Hplc i need to get Alpha tocopherol separately which has to match with Alpha tocopherol reference, ie same Retention time. Whether any special method is there for estimating alpha tocopherol from Alpha tocopheryl acetate
Use a C18 reverse phase column and an isocratic mobile phase comprised of 95% Methanol and 5% Water. If the flow is 0.5 mL/minute with a 250 mm column the D-a-tocopherol will elute around 11-12 minutes and Tocopheryl Acetate about a minute later. These peaks are baseline resolved.
As for converting the Vitamin E Acetate to IU equivalents of Vitamin E I would use a standard table (many are online). Try a 'Google' search.
