While doing synthesis using hyaluronic acid we found unreacted HA (10KDa) still remaining after dialysis (14 Kda MW cut-off). Can you suggest any explanations or ideas?
The dialysis has to be performed in 100 mM NaCl as then it will remain in the carboxylate state. It is unlikely to get HA of 10 KDa left after doing dialysis in a 14KDa bag. Dialysis was performed for how much time? Which dialysis bag did you use? Does it require any pre-activation? Dialysis has to be done for 3 days to get everything out. Use 100 mM NaCl for 1st and 2nd day, and on the 3rd day do in plain water. If you expect any amine byproduct in you reaction, perform dialysis in pH 3.5 (add 3 drops of conc HCl in 2 L water) for the first 2 days. Hope it will help.
