In HPLC-DAD there is always persistent peak that comes very early and with a valuable Absorbance even with blank samples may be it comes from the solvent or so, How could I get rid of it in order to not appear in the final chromatograme ?
I can only erase the integration line to not counting it as a peak area but still I need to remove it at all from the graph .. is there something like background correction or so? I understand mostly it comes from what so called volume void time
You can dissolve your compound in your mobile phase instead of other solvents and then inject it into into the HPLC.
As noted, first, please make sure you are following good chromatography fundamentals and dissolving your sample in the mobile phase (not a stronger solvent). That should help improve chromatography peak shape and result in a smaller initial peak.
Next, how do you know that the peak you observed is from your mobile phase? Have you developed a proper method which retains, holds, then elutes ALL materials ? Did you look at it's UV/VIS spectra ? Start with the basics that all chromatographers should know. What is the column void void for your method (measured and calculated)? The T zero time ? What are the K primes for each peak? Do these values imply that the method follows good practices? Finally, I like to see a peak at the beginning of the analysis showing the injection pulse. It is useful in confirming the actual void volume and shows anyone with a basic understanding of liquid chromatography a critical troubleshooting reference value.
BTW: That "peak" should not interfere with any of your calculations and if you have your current integration events / parameters set up to record the peak, then you can easily add an event to the Integration Table to either ignore it or to not start integrating peaks until after it has appeared. Just make the change, and save it as part of your data analysis method.
Thank you a lot , that was so helpful , now I became convened that this peak due to the T zero , I have done the calculated and the measured T zero and they are the same also when I used 0.5 flow rate It became half
