I have screen some compounds from plants for lipase inhibitors. I have to use high through put screening but plant pigments disturbs the high through put screening method. I want to know the method how to remove pigments?
SPE on RP-18 could be a solution, but this will also remove some lipophilic secondary metabolites. Not sure about the elution solvent strenght, I would try Methanol-Water (7:3).
The need for a method compatible with-throughput screening may exclude or limit the possibilities of using SPE/other resins. Without spending bigger resources in this case, it might be better to perform a simple liquid extraction, which may not allow to isolate a whole set of lipase inhibitors but only a subset: In case your pigments are water soluble, try to extract with chlorophorm or ether to find a subset of lipase inhibitors which are lipophilic. In chase your pigments are fat soluble, extract a subset of lipase inhibitors using polar extraction solvents. Good Luck!
