I am trying to quantify the number of cells in the liquid fermentation. The liquid fermentation including CaCO3 which makes it impossible to do quantification by UV-OD600 and weigh the dry weight directly. I am thinking to use HCl to get rid of CaCO3 at first. Just wondering will the HCl influence the weight of cells and do you have any good idea to quantify the cells when your fermentation containing CaCO3?
Hello, you can try to keep the tubes under 4"C than perform decantation of upper liquid or better to count the cells by surface playing after series of decimal dilutions.
Also take in to count that to avoid this problem in future better to use CaCo3 tablets not powder which can be easily removed after fermentation.
How about using a weaker acid than HCl to reduce the influence of a low pH on cell protein stability or even osmotic problems that reduces cell size? Once CaCO3 dissolves in acetic acid pH keeps near to neutrality preserving cell structure. Also, if CaCO3 amount is little, injecting CO2 might solubilize it into calcium bicarbonate; it might work too...
Complexation of Ca (and Mg) using an EDTA Containing buffer. Of cause ghis is only an option if Ca concentration is not too high.
