Actually i want suggestions from the experts that how to quantify amino acids from a plant sample.
I'll be highly indebted if anybody is here to answer in an elaborated way. Keep in mind i have yet the powdered plant sample only.

Tawseef Ahmad Mir: Please try a keyword search on the web for more information (e.g. Google. amino acid analysis by HPLC, plant). Investing your own time to learn about the techniques and methods used before moving forward with any project of this complexity is key to success. This web forum is not the place to teach a complex class on amino acid analysis by HPLC (no one can teach you the years worth of knowledge needed in a day, week, month etc). Amino acid analysis by HPLC is a complex form of analysis and best suited to the most experienced liquid chromatographers (requires a great deal of practical experience, knowledge and training to achieve accurate results). This is not a good application for a student to do on their own. Please enlist the help of a professional chromatographer or laboratory to assist you with this project. You will save a great deal of time in this way and learn more about the process.

Amino acid profiles were analyzed after hydrolyzing the samples with 6 N hydrochloric acid in a sealed tube for 22 h at 110 °C in a vacuum oven. The acid was evaporated using a vacuum rotary evaporator and the residue was placed in a diluent (0.1 N hydrochloric acid) and then filtered using 0.2 μm membrane syringe filter. Amino acids were qualified and quantified by a fluorescent detector (RF-20AXS) using the amino acid mixer as an external standard. Tryptophan, being liable to acid hydrolysis, was measured after alkali hydrolysis by spectrophotometric method at 500 nm. The partial oxidation of methionine during acid digestion was prevented by using 0.1% phenol.
