To make PHB standard, I used 4 mg PHB from Sigma, 2 ml of methanol containing 7% (v/v) concentrated H2SO4 and 4 ml of chloroform. i heated it at 100 degree C for 2.5 hours, then added 4 mL water. I took the bottom phase (chloroform part) and dried in in sodium sufate before for GC analysis, but there was no peak for methyl ester of PHB. which step is wrong?

Sometimes I found that the color of mix solution after heating is change to be red/brown and the solvent is reduced, but it can not be dissloved in the chloroform. is it methyl ester of PHB?
