hi guys, i need to predict, given a structural protein back bone what are the position that can lead to a disulfide bound. i need it cause my purpose is to insert a disulfide bridge in a protein and i need to know the most appropriate site where mutate for the cysteines. Thank you.

I understand that you are designing a protein for recombinant expression, and want to introduce a disulfide bridge to change or improve function? If so, the best option is to model the protein for homology using PDB templates, insert the corresponding disulfide bridge, and then to evaluate if the protein is thermodynamically stable.
