I am trying to prepare PHA samples for the GC-MS analysis, during the procedure PHA is mixed with an equal volume of acidified methanol and chloroform. The latter is boiled at 100 degrees Celsius for 2 hours. After the digestion distilled water is added which tends to separate out the organic phase. However, during boiling, the content chloroform evaporated even after tight capping and after the addition of distilled water no organic layer is formed.
Is there any other method to prepare the samples for GC-MS? What can be done to modifiy the conventional method?
