Through field water balance approach one can work out the periodic water use (in terms of evapo-transpiration), which you can add for the entire growing season. Then one can divide biomass (or economic yield) by the seasonal water use for getting water use efficiency.
There are crop simulation models as DSSAT, INFOCROP, APSIM which work out the water use (evapo-transpiration) as well the grain yield and one can get the water use efficiency.
Water use efficiency is not conservative, it varies depending upon the climatic extremes, nutrients & water application inputs
I have carried out lot of work on this aspect

Water use efficiency of crop can be measured if the biomass (economic use) of the crop and seasonal consumptive water use are known. Biomass divided by consumptive water use during the growing period provides data on water use efficiency
The FAO Acquacrop model can help you in water efficiency estimation. >>>For direct measurement there are differnt ways, raported in theoprevious post.

The term "water use efficiency" is a very broad term, and can have different meanings according to the specific interest of the researcher or manager. In Australia there was much discussion some years ago on this issue, and the consensus which emerged was that you really need to define your term clearly, whatever term you choose to use.
