hi, I want to measure adhesion force of cell with substrate. I am not accesible to AFM and other force measuring instrument. is there any protocol to measure force using micropipette and standard microscope.
You may be able to pull a long pipette tip that would allow you to do a deflection calibration on an analytical balance to obtain the spring constant and then use the pipette to dislodge the cell from it's adhesion point while measuring the deflection either in the microscope or with a micrometer. A student in our lab used this method to calibrate PDMS post arrays for measuring cell adhesion/traction forces. http://etd.library.vanderbilt.edu/available/etd-08182008-153315/unrestricted/KwekuAddaeMensah_Diss20080818.pdf
