To determine MIC using 96 well plate, if the synthetic compound is only soluble in DMSO, then how to make two fold serial dilution of 5mg dissolved in one ml pure DMSO ?
you can dilute your 5 mg/ml stock solution directly in the medium used for the MIC assay. It should not precipitate as you go down a bit with the concentration. Anyway you can check it before doing the actual assay. If needed you could also do a pre-dilution in 50% DMSO and then go in your medium.
I agree with Francesco. I would simply suggest you to dilute your stock solution directly in the medium, since DMSO is toxic per se, and it could hide the real MIC. Similarly, it would be useful if you could prepare a more concentrated stock dilution, so that you can serially dilute it, preferentially with the testing medium...
Anyway, take care to the pH of the solution, it could lead molecule precipitation even if it is dissolved in DMSO... but you should easily notice it at a glance.
