Sometimes I see comments which are added in the end of a paper(e.g. I've seen comments and their replies from the author in the end of the paper in Elsevier's journals). The question is that how can we make such a comments to get answer from the author(s) formally?
I have several questions on a paper that should ask them to the authors. I've tried to have a conversation with the authors via E-Mail but nobody answered me then decided to ask my questions formally from the journal. May I use LTE (Letter to the editor) to do this? have you ever done such a thing so far?

It is hard. My questions have never been answered. But with a little effort, you can usually work it out yourself
Letters to the Editors make sense only when the journal accepts and publishes these. (See instructions for authors of the specific journal.) And even when it does there is no guarantee that your letter will be published. Editors may reject letters as they reject submitted papers.
As Wolfgang R. Dick notes, there is considerable variation from journal to journal about whether they take comments and whether proposed comments are reviewed or just decided by the editor. I think the easiest way to determine if a journal accepts comments is to look at back issues.
