I'm doing a enzymatic reaction that needs to be done in anaerobic condition. First step is to equilibriate proteins and reagents in the glove box with N2 to get rid of any remaining O2. The glove box is at room temperature. I want to keep my protein cold when equilibriating. Does anyone have suggestions on how to make a cold box or something like that inside of the glove box?
Dear Peng Ge , my first idea would be to use a cold bath, or just keeping the reaction vessel in dry ice. However these two simple approaches might interfere in some way with your equilibration with N2, because some other gases could be introduced inside the globe box. You could use a liquid N2 bath as well but also a good option -depending on the temperature range you want to keep your reaction- would be to rely on a thermoelectric cooling using a Peltier cell given that kind of system is not introducing any kind of gas or vapour into the chamber and it works efficiently for a lab scale reaction volume.
