I am trying to load Hesperidin on Gold nanoparticles. I am using already prepared Gold nano purchased from Sigma Aldrich. I tired so many times to conjugate gold nano with Hesperidin but failed to do so. Then I tried GSH as a ligand and put GSH(glutathione) in already prepared Gold nanoparticles (size 5nm OD 1, stabilized suspension in 0.1 mM PBS, reactant free) and stired it for 30 minutes. Then I dissolve Hesperidin in DMSO and mix this Hesperidin with Gold nanoparticles and GSH solution and kept this solution altogether for overnight continuous stirring at room temperature. Then this solution was ultracentrifugated at 13000rpm for 30 minutes to remove excess Hesperidin. Then I performed FTIR which now showed functional groups. Now I am confused that GSH is also an antioxidant just like Hesperidin. Can anyone help me answer this question that as I mentioned FTIR showed the conjugation of Hesperidin with gold Nanoparticles was it actually conjugated together and GSH worked as ligand or it was GSH which was conjugated. How can I confirm the conjugation?
