Hi everyone,

I'm working with a double stranded DNA sequence that has a single stranded tail. The single stranded part can form strong hairpin structures (delG ~ -8), which I need to get rid of.

The double stranded part should have a melting temperature of ~80C, whereas Tm for the hairpin structures are ranging from 45-61C. I tried to heat the sample at 65C with formamide, but it seems to melt both the dsDNA and hairpin.

Is there a way to keep the single stranded part stay linear, without denature the double stranded section?
