Hi!

I'm conducting researching a project on MBsome(midbody).

I am interested in the midbody after seeing research paper (Understanding Post-Mitotic Roles of the Midbody during Cell Differentiation and Polarization) that isolates the midbody through sucrose density gradient, and proceeded in the same way with 40% glycerol cushion, 2M sucrose, and 1M sucrose. However, I expected the 40% glycerol cushion to be the heaviest.

But my tests show that 2M sucrose, 1M sucrose, and 40% glycerol are in order from the bottom layer, so there is no band formation between 2M and glycerol as in the paper.

Should glycerol be dissolved in powder form with sucrose buffer and diluted with water? Do you know a solution to this problem? Or, if there is an alternative to the midbody isolation method, please recommend it.
