I'm producing Cellulase using Aspergillus Niger according to Mandels and Weber, 1969.
Should i separate the enzyme from production media before using it to degrade lignocellulose materials particularly sugarcane bagasse? or can i use it as crude enzyme?
Yes it can be used as crude enzyme, and may produce good results, But concentration of enzymes may be required to get better results.
After the production of enymes using production medium used by Mandels and Weber (1969) it can directly be used as crude enzyme preparation to determine the various cellulolytic activities. But, the fungal mycelium must be separated using centifugation followed by filteration.
The enzymes (protein) are present as soluble form (dissolved in the solvent of your production medium, distilled water). To saperate the enzymes/total protein, you must precepitate out your protein by any precipitation method (like TCA/acetone/phosphate) which can also be taken as concentration method of your total protein fold.
The concentration method will also depend on the nature of your final use of enzyme where you want to use them and also the sensitivity of the enzymes towards the concentration method, as it must not be interfering with the activity of the enzymes.
