Dear all, recently I want to check the effect of an antifungal compound on the mycelial growth of Cryptococcus neoformans. I knew that most of the researcher induced them by mating them on V8 medium. May I know is there any other method for me to use in order to induce mycelial growth of Cryptococcus neoformans other than mating them? Could I just incubate them in RPMI-1640 medium? Thank you.
Dear Phil Geis, thank you for your answer. The paper is very useful. However, I do not understand what is the "filament agar" mentioned in the paper by the author. They used a lot of medium, YPD, MIN agar, V8 medium and filament agar. But, I do not know what is filament agar.
For your question, the antifungal compound I used is against pathogenic fungi by inhibiting the hyphal growth of the fungi. That is why I want to know what will happen if they are applied on cryptococcus neoformans if they are in the hyphal form.
Their ref's 6,7,8 apparently define "filament agar." Have you been able to track those down? If those aren't useful or you can't access, suggest you email the corresponding author at [email protected].
My question - the hyphal form of C. neoformans is not associated with human disease. As the parasitic form is yeast-like and the morphologies are so divergent, what would be the significance of hyphal susceptibility?

Dear Phil Geis, thank you for your help. I did read the papers cited by the author but those papers do not mention clearly about the filament agar. But, I have sent the email to them and they already replied me regarding to the filament agar. For your question, I understood your question. Actually, I was asked by my supervisor to check the effect of the antifungal compound on Cryptococcus neoformans hyphal growth even though we knew that they invade human tissue in yeast form. I think that, he wants to know its effect on basidiomycetes hyphal growth and we do not have much basidiomycetes in our laboratory. So, Cryptococcus neoformans is the only choice for us, may be. But, of course this fungi is not our first priority in my research. But, he just want to know if the drug exhibits inhibitory effect on basidiomycetes hyphal growth regardless of their significance in the research. By the way, I have found another paper which might be more useful.

This paper showed that the mutant of Cryptococcus neoformans could form hyphae in certain liquid medium, without mating, instead of on the agar.
