I'm trying to induce apoptosis in hemocytes in suspension from bivalves. For detection, I use the CellEvent™ Caspase-3/7 Green Flow Cytometry Assay Kit from ThermoFisher to mark apoptotic cells and Sytox to mark dead cells. I have tried paraquat, copper and rotenone to induce apoptosis and i'm not able to increase apoptosis with these products. I'm wondering wether I'm using the wrong agents to induce apoptosis or the right fluorescent probes.
I would glad if some of you could share there experience

Gervais et al. 2015 used dexamethasone (10 µM final concentration) to induce apoptosis in hemocytes of the flat oyster (i.e. as a positive control for the Vybrant FAM Poly Caspases Assay Kit, Molecular Probes).
Interestingly, caspase activation was similar in control and in hemocytes treated with dexamethasone. In their study, UV exposure caused caspase activation in oysters from Quiberon, but not in oysters from Corsica.
